Any idea what kind of speed I could get from a SATA III Raid Card which is plugged into my PCI-Express? Since this MB has trooped along, I would like to get the best speed possible without trashing a reliable MB like this one.
Motherboard: GA-EP45-UD3R/UD3
The SSD I am looking at is:
Crucial M4 2.5 inch 64GB SATA3 (x2 Raid 0)
or a single
Crucial M4 2.5 inch 128GB SATA3
Raid Card:
HighPoint Rocket 620-OEM PCI-Express 2.0 SATA III (6.0Gb/s)
Side Question.... Should I go with 2 64GB's in Raid or a single. Can I run a single SSD on the Raid Card?

Yes,Yes, Yes and let me think!................Yes..... as long as you have at least 4X PCI-E slot available, any of the kit you stated will run at or near the spec speeds, and yes you can boot of this SATA III spec raid card...........the hard thing to do IS NOT boot off it.....believe me!...
as this card has two raid controllers, best bang for buck is achieved with 2 not 4 drives hanging off it.....one for each controller
