Greetings,
Memory requirements differ based on the CPU. Summit, Raven or Pinnacle Ridge
Dual Channel mode is recommended, that's not to say the system couldn't boot (with one stick) in
some circumstances, but I wouldn't waste time trying to figure out what works and what doesn't. It's not designed for this.
Memory should be matched (brand, size and spec) and should be on the QVL for best results.
For reference:

This motherboard provides four memory sockets and supports Dual Channel Technology. After the memory is installed, the BIOS will automatically detect the specifications and capacity of the memory. Enabling Dual Channel memory mode will double the original memory bandwidth.
The four memory sockets are divided into two channels and each channel has two memory sockets as following:
Channel A: DDR4_2, DDR4_4
Channel B: DDR4_1, DDR4_3
Due to CPU limitations, read the following guidelines before installing the memory in Dual Channel mode.
1. Dual Channel mode cannot be enabled if only one memory module is installed.
2. When enabling Dual Channel mode with two or four memory modules, it is recommended that memory of the same capacity, brand, speed, and chips be used. For optimum performance, when enabling Dual Channel mode with two memory modules, we recommend that you install them in the DDR4_1 and DDR4_2 sockets.
Recommend that you populate 2 slots and
use QVL RAMOn an aside. 1 long beep with 3 short. That's No VGA detected. 1 long and 2 short is the same. Solve your RAM issue first and see if this remains.
