Dear Customer,

If the drive does contain an bootable OS it should not be an issue booting off from it. Make sure that hdd boot priority under bios is configure as the drive as the first order and first boot device as USB hdd. Connect it onto the rear usb ports off the back of the board

Intel Core i5-2400 Sandy Bridge 3.1GHz.
G.SKILL Ripjaws X Series 4GB (2 x 2GB) 240-Pin DDR3 SDRAM DDR3 1600.

ASUS CD/DVD DRW-24B1ST SATA 24.
Corsair PSU CX600.
Cooler Master Elite case.

Yes, from GGTS. I feel pretty stupid but I should have known that running a USB pendrive boot works best from a rear USB port.  It does work now. I'm sorry for my confusion and ranting. All is well now.   ;)

To be fair I didn't think to mention it myself which I guess I should have, I just assumed that you were using a main USB port. Anyway at least it's all working now. ;)

Thank you once again. It is odd that  Iliaspap here disabled  Legacy support of USB USB mass storage and it seemed to fix his problem. I have 3 pendrives that I've booted on other machines but none of them work consistently with this motherboard. I will see shortly if my new USB 3.0 pendrive will work when it arrives.

After 3 days, my system is rock stable, no problems anymore, I have my usb 2.0 WD connected on the back usb ports, tested both usb2 and usb3 ports, same resaults. Also I didnt disabled USB Legacy support, I disabled only the USB mass Storage Fuction. And to be certain, I just tested to re-enable usb mass storage function, I had the same issues again, Slow Bios Post and Took ages to load windows7 x64. So It is 100% the solution of my problem.
