HI and welcome to the forum.
I am a little bit confused by what is/or isn't happening here.
Do you have speakers plugged into the front jack output?

Ok sorry let me explain my problem a bit more

I have a GIGABYTE GA-EP45-DS3R Motherboard inside a Cooler Master Elite 330 Chassis (http://www.coolermaster.com/product.php?product_id=30&category_id=19 (http://www.coolermaster.com/product.php?product_id=30&category_id=19))

The chassis' front audio panel supports AC97 audio only while the motherboard is HD

I plugged everything in properly following both manuals.

I installed all drivers and Realtek Audio Manager.

But when i try to plug in speakers or headphones no sound comes out.

Any suggestions?

You mean no sound also when connecting the speakers to the rear ports on the motherboard?

If so please try installing the latest Realtek drivers from here
http://www.realtek.com.tw/downloads/downloadsCheck.aspx?Langid=1&PNid=14&PFid=24&Level=4&Conn=3&DownTypeID=3&GetDown=false

Also, open Control panel, sounds and be sure the HD sound device is enabled.   All my comments are in regards to testing your speaker connected to the rear HD audio ports.

The motherboard is compatible with either HD or AC97 audio to the front panel.
I took this pin out  and extra information from the manual

For AC'97 Front Panel Audio:
• The front panel audio header supports HD audio by default. If your chassis provides an
AC'97 front panel audio module, refer to the instructions on how to activate AC'97 functioninality
via the audio software in Chapter 5, "Configuring 2/4/5.1/7.1-Channel Audio."
• Audio signals will be present on both of the front and back panel audio connections
simultaneously. If you want to mute the back panel audio (only supported when using an HD
front panel audio module), refer to Chapter 5, "Configuring 2/4/5.1/7.1-Channel Audio."
• Some chassis provide a front panel audio module that has separated connectors on each
wire instead of a single plug. For information about connecting the front panel audio
module that has different wire assignments, please contact the chassis manufacturer.

No i'm connecting the speakers to the front because it's really just a cd player connecting to my computer with an audio cable. I also already have the latest Realtek Drivers

I already read that part of the manual and activated ac97 but if i plug in my headphones the only time i hear sound on my headphones is when i press the play button on the realtek panel

Do you have the CD IN cable connected to the motherboard then?  Is that what you are trying, to listen to a CD/DVD from the player?

Do you hear windows sounds, test with control panel, sounds, then right click your speakers and choose test.   You may need to change the configuration on them, try all options 2/2.1/4.1/ect and see if any work.
